Brief Introduction

Penciclovir is a guanine analogue antiviral used to treat various herpes virus infections. It is a nucleoside analogue with low toxicity and good selectivity. Penciclovir is a selective anti herpesvirus drug, especially for herpes simplex virus type I and type II and varicella zoster virus. Penciclovir is converted into its active form by cell kinase, penciclovir triphosphate. It competitively inhibits viral DNA polymerase with deoxyguanosine triphosphate. It can inhibit the DNA synthesis of virus infected cells. Penciclovir has in vitro activity against herpes simplex virus type I and type II, but does not inhibit DNA synthesis in uninfected cells. The antiviral effect is exerted by the qualitative change of viral thymidine kinase or DNA polymerase. Studies on herpes simplex virus type I have shown that penciclovir can induce apoptosis without causing too much genotoxicity.
